丂Accumulating evidence now implicates fallopian tube secretory epithelial cells (FTSEC) as precursors to serous high-grade epithelial ovarian cancers (OVCA). Serous tubal intraepithelial carcinomas (STICs), which are associated with p53 signatures and DNA damage, are precursor lesions derived from FTSECs. We propose that additional alterations mediated by chronic stressors such as iron-induced oxidative stress may promote the transition of precursors to high-grade serous OVCA. Specifically, pelvic inflammation or iron overload disease may exacerbate the initial DNA damaging events. A causative relationship between iron exposure and ovarian cancer risk has not yet been explored experimentally and is our current area of investigation. We now show that chronic iron exposure of FTSECs increased their proliferative and migratory indices. Further, we identified that chronic iron exposure altered expression of Ecotropic Viral Integration Site-1 (EVI1, located at 3q26.2, a notable amplified region in serous high-grade OVCA), which upregulated telomerase reverse transcriptase (TERT) expression. Current work focuses on the link between EVI1 and the sphingolipid axis to the pathogenesis of OVCA.

My studies presently aim to answer : What are the structural proteins of the mitotic chromosome and how do they direct chromosome segregation in mitosis? Currently, we have identified ~4,000 polypeptides in highly purified chicken mitotic chromosomes by using a mass-spectrometry based proteomic analysis. This seminar is going to focus on a novel approach that we term Multi-Classifier Combinatorial Proteomics, to sort through this massive proteomic data set and integrate multiple classifiers by machine learning uncovered functional relationships between protein complexes in the context of intact chromosomes.
